Dоsage forms with modified release: advantages, disadvantages and some problems associated with them

Modified-release dosage forms are those which release the drug substances slowly. They can be used to reduce the number of admissions, maintain constant plasma concentration levels. They are convenient to take and administer. They are also preferred in cases of polymorbidity, in which more than one dosage form is taken daily. Avoiding taking at night and unwanted fluctuations in plasma concentrations are avoided. The help optimise the action of well-established medicinal substances and reduce side effects. By modifying the dosage forms, the disadvantages of classical ones, such as rapid release of the active substance, large differences in minimum and maximum plasma concentrations, undesirable side effects could be avoided. A review by the European Medicines Agency of medicinal products containing modified-release paracetamol was carried out in relation to the risks of complex release of paracetamol from these forms in the body in case of overdose.
